Don't put them directly on the glass, or you'll get what's called Newton Rings. They are caused by interference patterns , I think from the single molecules of air inbetween. Anyway, you will get concentric rings in places around the image, really annoying!
I've made thin cardboard or cardstock holders for odd sizes. Some scanners have a calibration area built into the film holders, that must be left open. I don't know about others than Epson on that score.
Full frame, 35mm slides, roughly 1" x 1 1/2" x 2400 ppi (pixels per inch) will give you prints @ 240ppi, about 10x15" which will look very good.
